What is the difference between a front loaded and a rear loaded amp cabinet?

Hi and welcome. This is an often overlooked subject. Frontloaded cabs sound tighter, more up front and crisper. Rearloaded sound like a more distant rumble, looser and less defined. Depends on your guitar/amp combo. I like more classic rock edgy tone so my preference is frontloaded, especially with my Herbert as it has gobs of lows anyway.

What is the physical difference in how they're actually built/set up? Is it quite literally just that the speakers are either loaded through the front or the back of the cabinet?

(I would take you up on the offer if you weren't on the opposite side of the planet. The cost of flight would be a good down payment on the amp! )

Just as the term suggests. The spekers are mounted on either the front or rear side of the face panel.
