For a guy that spends 80% of the time playing instrumental (solo) and/or lead guitar (band), what are some thing to keep in mind, or advice on setting hcannels?

Lead tones I like smooth, fat, midrangey!

rythms I like loud, well mannered meanness.

Cleans I like...any way! Ill be using my volume for most cleaner parts anyhow.

Which mode (if any) should I use plexi mode for?

For mid-range plexi style bite and bark.

My experiences after 4 years with that amp (I'm lead guitarist in rock/metal band) are:
- =C='s for poweramp - if it comes with JJ's put them to trash can immediately
- keep both boosts on or remove dirty boost bypass cap if You need drop in gain for dirty channels
- Structure High
- for dirty channels keep Middle at 2 o'clock or more (this is essential for trying to avoid "blanket effect")
- keep Presence at noon or more
- Excursion M for dirty channels
- assign second Presence to Ch. 1 and keep Excursion at L position
- play with pre-eq's but more likely You'll like B1 for both clean and dirty channels (especially dirty's, they sound lifeless and gainless with B2 4n7 cap)
- rest to taste

Plexi mode in 101B is useless IMO.

If You have any more questions regarding this amp feel free to ask.

Regards, Andy :)
